The MEPCE protein also known as 3-methylcytosine-specific methyltransferase plays an important mechanical role in RNA processing. MEPCE has a mass of approximately 81 kDa. It adds a methyl group to the 7SK snRNA a critical step in the regulation of transcription. This protein is expressed across various tissues indicating its important functions in different cellular environments.
Biological function summary

The MEPCE protein is an important player in regulating gene expression by stabilizing the 7SK snRNA in a specific ribonucleoprotein complex. This complex controls the activity of the positive transcription elongation factor b (P-TEFb) an important factor in the transcription elongation stage of gene expression. By methylating 7SK snRNA it helps maintain the repression of P-TEFb balancing transcriptional activities.

Pathways

MEPCE works collaboratively within the RNA polymerase II transcription machinery. It participates in the regulation of transcription elongation an important pathway in gene expression. MEPCE interacts closely with other proteins in this process such as LARP7 and HEXIM1 which also associate with the 7SK snRNA to modulate P-TEFb activity.

MEPCE's role in transcription regulation links it to various regulatory diseases including certain cancers and cardiac hypertrophy. The imbalance in P-TEFb activity which MEPCE helps control can lead to unregulated cell proliferation or heart muscle enlargement. MEPCE in association with LARP7 and HEXIM1 adds a layer of complexity to these conditions as alterations in this regulatory system can provoke disease onset or progression.

S-adenosyl-L-methionine-dependent methyltransferase that adds a methylphosphate cap at the 5'-end of 7SK snRNA (7SK RNA), leading to stabilize it (PubMed : 17643375, PubMed : 19906723, PubMed : 30559425). Also has a non-enzymatic function as part of the 7SK RNP complex : the 7SK RNP complex sequesters the positive transcription elongation factor b (P-TEFb) in a large inactive 7SK RNP complex preventing RNA polymerase II phosphorylation and subsequent transcriptional elongation (PubMed : 17643375). The 7SK RNP complex also promotes snRNA gene transcription by RNA polymerase II via interaction with the little elongation complex (LEC) (PubMed : 28254838). In the 7SK RNP complex, MEPCE is required to stabilize 7SK RNA and facilitate the assembly of 7SK RNP complex (PubMed : 19906723, PubMed : 38100593). MEPCE has a non-enzymatic function in the 7SK RNP complex; interaction with LARP7 within the 7SK RNP complex occluding its catalytic center (PubMed : 19906723).
