Immunofluorescent analysis of 4% Paraformaldehyde-fixed, 0.1% TritonX-100 permeabilized 293T (human embryonic kidney epithelial cell) transfected with a MERS-CoV Nucleocapsid protein expression vector containing a myc tag. cells labelling MERS-Cov Nucleocapsid protein with ab316332 at 1/200 (5.08 ug/ml) dilution, followed by Goat anti-Human IgG (H+L) Cross-Adsorbed Secondary Antibody (Alexa Fluor® 488) antibody at 1/100 (5ug/ml) dilution (Green).
Confocal image showing cytoplasmic staining in 293T cells transfected with MERS-CoV Nucleocapsid expression vector containing a Myc-his-tag® (shown in green). The counterstain was observed in red. Nuclear DNA was labeled with DAPI (shown in blue).
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
ab223895 Anti-Myc tag Mouse monoclonal antibody (Alexa Fluor® 647) was used to counterstain at 1/100 (5ug/ml) dilution (Red). The Nuclear counterstain was DAPI (Blue).
Secondary antibody only control : Secondary antibody is Goat anti-Human IgG (H+L) Cross-Adsorbed Secondary Antibody (Alexa Fluor® 488) at 1/100 (5ug/ml) dilution.

Biological function summary
The MERS-CoV Nucleocapsid protein interacts closely with other viral proteins including the membrane (M) protein to ensure successful viral replication and assembly. This protein does not work alone; it is part of the larger ribonucleoprotein complex which plays an essential role in stabilizing the viral RNA. Its interaction with the M protein helps in localizing the N protein to specific sites in the host cell preparing the components necessary for virion assembly and release.
Pathways
The MERS-CoV Nucleocapsid protein engages in processes important for viral pathogenesis and host cell interaction. The protein is part of the coronavirus replication and transcription complex pathway. Within this matrix it associates with non-structural proteins like the RNA-dependent RNA polymerase to enhance RNA synthesis. Additionally it partakes in modulation of host processes to evade the immune response a pivotal aspect of how coronaviruses maintain infection and propagate within the host.
