Supplementary information

METTL21A also known as FAM119A is a methyltransferase enzyme involved in protein methylation. It belongs to the METTL (methyltransferase-like) family and has a molecular weight of approximately 28 kDa. METTL21A is ubiquitously expressed in various tissues with relatively higher levels in the kidney and liver. It acts through catalyzing the transfer of methyl groups to target proteins often modifying lysine residues which can influence protein function and stability.
Biological function summary

METTL21A functions in protein post-translational modification and plays a role in regulating protein interactions and signaling pathways. Although not strictly part of a specific complex METTL21A interacts with substrates and partner proteins affecting cellular function. The enzyme's activity can influence cellular processes such as signaling growth and apoptosis which are essential for normal cellular homeostasis.

Pathways

METTL21A participates in the modulation of signaling pathways like the mTOR pathway. This pathway is important for cell growth and metabolism wherein METTL21A may impact its regulation through methylation of pathway-related proteins. Relationships with other methyltransferases or signaling proteins such as those in the S6K family demonstrate its significance in controlling biological responses to nutrients and growth factors.

METTL21A associates with metabolic diseases and certain cancers. Its altered expression or function can contribute to abnormal protein methylation which is evident in some forms of cancer possibly affecting tumor growth and progression. Furthermore METTL21A's influence on metabolic pathways implicates it in disorders related to metabolic dysregulation potentially intersecting with proteins like mTOR and AMPK that are key players in cellular energy homeostasis.

Target data

Protein-lysine methyltransferase that selectively trimethylates residues in heat shock protein 70 (HSP70) family members. Contributes to the in vivo trimethylation of Lys residues in HSPA1 and HSPA8. In vitro methylates 'Lys-561' in HSPA1, 'Lys-564' in HSPA2, 'Lys-585' in HSPA5, 'Lys-563' in HSPA6 and 'Lys-561' in HSPA8.
