Mitochondrial S-adenosyl-L-methionine-dependent methyltransferase that mediates N(3)-methylcytidine modification of residue 32 of the tRNA anticodon loop of mitochondrial tRNA(Ser)(UCN) and tRNA(Thr) (PubMed:34774131, PubMed:35017528). N(3)-methylcytidine methylation modification regulates mitochondrial translation efficiency and is required for activity of the respiratory chain (PubMed:34774131, PubMed:35017528). N(3)-methylcytidine methylation of mitochondrial tRNA(Ser)(UCN) requires the formation of N(6)-dimethylallyladenosine(37) (i6A37) by TRIT1 as prerequisite (PubMed:34774131, PubMed:35017528). May also mediate N(3)-methylcytidine modification of mRNAs (PubMed:28655767). The existence of N(3)-methylcytidine modification on mRNAs is however unclear, and additional evidences are required to confirm the role of the N(3)-methylcytidine-specific mRNA methyltransferase activity of METTL8 in vivo (PubMed:33313824, PubMed:34774131).

METTL8 also known as Methyltransferase-like protein 8 is an enzyme that catalyzes the methylation of RNA molecules. It belongs to the larger family of methyltransferases which are essential for various methylation processes. METTL8 has a molecular mass of approximately 67 kDa. The expression of METTL8 is observed in multiple tissues but it shows higher levels in the brain and kidneys indicating a potential role in the regulation of processes specific to these organs.
METTL8 participates in the regulation of RNA stability and processing. It operates as part of a complex that modulates RNA modifications which can affect gene expression patterns. The methylation activity of METTL8 influences RNA metabolism having potential implications on the stability of transcripts and the overall efficiency of translation.
METTL8 involvement in RNA processing pathways becomes evident. It plays a role within the RNA methylation pathway by interacting with other methyltransferases like METTL3. This interaction suggests that METTL8 might have a function in the N6-methyladenosine (m6A) methylation pathway that modifies the nucleobases within RNA molecules to regulate gene expression. The significance of these pathways involves the control of diverse cellular processes including development and response to cellular stress.
Abnormalities in METTL8 function have potential links to neurological disorders and certain cancers. In particular altered expression levels or mutations in METTL8 may contribute to neurodegenerative diseases correlating with aberrant RNA methylation patterns. Its role in cancer comes from the association with dysregulated methylation often seen in tumorigenesis. METTL3 a related protein also connects to these diseases highlighting the importance of the methylation network in maintaining cellular homeostasis and its disruption in disease states.
