Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-MEX3A antibody (AB79046)
Immunohistochemical analysis of paraffin-embedded human testis tissue using anti-RKHD4 antibody (ab79046) at 1 µg/ml. Tissue was fixed with formaldehyde and blocked with 10% serum for 1 h at RT; antigen retrieval was by heat mediation with a citrate buffer (pH6). Samples were incubated with primary antibody overnight at 4°C. A goat anti-rabbit IgG H&L (HRP) at 1/250 was used as secondary. Counter stained with Hematoxylin.

MEX3A is highly expressed in PDA and the construction of MEX3A knockdown cell model. a Expression levels of MEX3A in PDA tumor tissues and adjacent normal skin tissues were detected by IHC staining. b Kaplan–Meier survival analysis MEX3A expression and overall survival of PDA. c MEX3A expression in HPDE6-C7, BxPc-3, PANC-1 and SW1990 cells was detected by qRT-PCR. d qRT-PCR was used to screen knockdown efficiency of MEX3A in shMEX3A-1, shMEX3A-2, and shMEX3A-3 groups. e Infection efficiency for PANC-1 and SW1990 cells was evaluated by expression of green fluorescent protein 72 h post-infection. f, g The specificity and validity of the lentivirus-mediated shRNA knockdown of MEX3A expression was verified by qRT-PCR (f) and western blot analysis (g). The data were presented as the mean ± SD (n = 3). *p < 0.05, **p < 0.01, ***p < 0.001

Biological function summary
The MEX3A protein plays roles in cell differentiation and proliferation by influencing mRNA dynamics. It operates independently rather than as a part of a larger protein complex. MEX3A regulates the expression of key developmental and cell cycle-related genes aiding in cellular function and tissue homeostasis. The protein's ability to modulate mRNA transcripts makes it a significant player in maintaining proper cellular response to environmental changes.
Pathways
Studies show that MEX3A plays a role in the Wnt signaling and Notch signaling pathways. In the Wnt signaling pathway it affects the stability of mRNAs related to cell growth and differentiation. Through the Notch signaling pathway MEX3A interacts with various proteins such as DVL3 influencing decisions in cell fate and proliferation. These pathways highlight MEX3A's involvement in important processes of development and cellular communication.
