MFAP4 also known as Microfibril-Associated Glycoprotein 4 is a protein that plays a significant role in the extracellular matrix architecture. The molecular mass of MFAP4 is about 29 kDa. This protein is expressed in various tissues including the lung liver heart and blood vessels. As a part of the extracellular matrix MFAP4 contributes to the structural integrity and elasticity of connective tissues.
Biological function summary

MFAP4 interacts with components of the microfibrillar network. It is not a standalone entity but functions as part of a larger complex that includes elastin and microfibrillin-1. This association enhances the mechanical properties of tissues maintaining them against pressure and stretch forces. MFAP4 also participates in cellular responses such as cell adhesion and migration which are critical for tissue remodeling and repair processes.

Pathways

MFAP4 engages in multiple biological pathways related to the extracellular matrix and tissue homeostasis. The TGF-beta signaling pathway is notable as MFAP4 interacts with multiple proteins within this pathway such as fibrillin-1. Through these interactions MFAP4 influences the regulation of cell growth and differentiation. Another pathway is the integrin signaling pathway where its association with integrins supports cell adhesion and communication within the extracellular matrix.

MFAP4 shows associations with fibrosis and chronic obstructive pulmonary disease (COPD). Both conditions involve alterations in the extracellular matrix composition and function. In fibrosis the dysregulated expression of MFAP4 can affect connective tissue remodeling. Altered MFAP4 levels have also been observed in COPD where it may impact lung tissue elasticity.
