Supplementary information

MFSD2A also known as Major Facilitator Superfamily Domain-Containing Protein 2A functions as a transporter protein. It efficiently facilitates transport of specific lipids across cellular membranes. This protein has a mass of approximately 56 kDa and is mainly expressed in the blood-brain barrier and endothelial cells. MFSD2A's expression in these areas plays an important role in transport through the protective barrier in the brain.
Biological function summary

MFSD2A is important in mediating uptake of omega-3 fatty acids such as docosahexaenoic acid (DHA) into the brain. It achieves this by ensuring proper distribution of lipids necessary for maintaining brain health and function. While not known to be a part of a larger complex MFSD2A works closely with lipid molecules and other membrane transporters to regulate lipid homeostasis within the brain.

Pathways

MFSD2A is involved in the lipid metabolic pathway and the neurovascular signaling pathway. Its activity significantly affects how lipids are transported and utilized within these systems. It interacts functionally with proteins like ABC transporters which also contribute to lipid processing and transport. These pathways highlight the importance of MFSD2A in lipid utilization and brain metabolism.

MFSD2A is closely linked to neurological conditions such as microcephaly and certain neurodevelopmental disorders. Its malfunction or altered expression can disrupt the supply of essential lipids to the brain leading to developmental abnormalities. The protein's role connects to other transporters like FABP5 hinting at a network of lipid transport mechanisms that when altered contribute to these diseases.

Target data

Sodium-dependent lysophosphatidylcholine (LPC) symporter, which plays an essential role for blood-brain barrier formation and function (PubMed : 24828040, PubMed : 32572202, PubMed : 34135507). Specifically expressed in endothelium of the blood-brain barrier of micro-vessels and transports LPC into the brain (By similarity). Transport of LPC is essential because it constitutes the major mechanism by which docosahexaenoic acid (DHA), an omega-3 fatty acid that is essential for normal brain growth and cognitive function, enters the brain (PubMed : 26005868, PubMed : 34135507). Transports LPC carrying long-chain fatty acids such LPC oleate and LPC palmitate with a minimum acyl chain length of 14 carbons (By similarity). Does not transport docosahexaenoic acid in unesterified fatty acid (By similarity). Specifically required for blood-brain barrier formation and function, probably by mediating lipid transport (By similarity). Not required for central nervous system vascular morphogenesis (By similarity). Acts as a transporter for tunicamycin, an inhibitor of asparagine-linked glycosylation (PubMed : 21677192). In placenta, acts as a receptor for ERVFRD-1/syncytin-2 and is required for trophoblast fusion (PubMed : 18988732, PubMed : 23177091).
