Immunofluorescent analysis of 4% Paraformaldehyde fixed 0.1% Triton X-100 permeabilized IMR-32 (human neuroblastoma neuroblast) cells labelling Mint-1 with ab190359 at 1/500 (1.964 μg/ml) followed by ab150081 Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) preadsorbed antibody as secondary at 1/1000 (2 μg/ml) dilution. Confocal image showing positive staining in IMR-32 cells (shown in green). ab7291 Anti-alpha Tubulin mouse monoclonal antibody at 1/1000 (1μg/ml) was used as a counterstain along with ab150120 as secondary at 1/1000 (2μg/ml) (shown in magenta). Negative control : HeLa. Nuclear DNA was labelled with DAPI (shown in blue). Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

Immunohistochemical analysis of paraffin-embedded human lung tissue labeling Mint-1 with ab190359 at 1/1000 dilution.

Low expression tissue : nearly no staining on human lung.

The section was incubated with ab190359 for 30 mins at room temperature.

The immunostaining was performed on a Leica Biosystems BOND® RX instrument.

Counterstained with Hematoxylin.

Heat mediated antigen retrieval was performed with Tris-EDTA buffer (pH 9.0, Epitope Retrieval Solution2) for 20 mins.

Putative function in synaptic vesicle exocytosis by binding to Munc18-1, an essential component of the synaptic vesicle exocytotic machinery. May modulate processing of the amyloid-beta precursor protein (APP) and hence formation of APP-beta. Component of the LIN-10-LIN-2-LIN-7 complex, which associates with the motor protein KIF17 to transport vesicles containing N-methyl-D-aspartate (NMDA) receptor subunit NR2B along microtubules (By similarity).
