Various whole cell extracts (30 μg) were separated by 7.5% SDS-PAGE, and the membrane was blotted with Miz1 antibody [HL1548] (ab308167) diluted at 1 : 1000. The HRP-conjugated anti-rabbit IgG antibody was used to detect the primary antibody, and the signal was developed with Trident ECL plus-Enhanced.

Miz1 also known as Zinc finger Miz1 is a transcription factor with a mass of around 87 kDa. It features multiple zinc finger domains allowing it to bind DNA directly. Miz1 is widely expressed in various tissues particularly in the nucleus of cells. Its function includes both activating and repressing gene transcription depending on the context. By modulating transcription Miz1 plays a critical role in cellular proliferation and differentiation.
Biological function summary

Miz1 interacts with other transcription factors and forms part of larger complexes that influence the expression of genes involved in cell cycle regulation. Frequently Miz1 associates with the Myc protein which modifies its activity allowing for tight control over its function. Through these interactions Miz1 regulates important genes that can tip the balance between cell survival and apoptosis.

Pathways

The function of Miz1 integrates into the TGF-beta and MAPK signaling pathways. In the TGF-beta pathway Miz1 collaborates with Smads to regulate target genes that govern cell growth. Transcription factor that can function as an activator or repressor depending on its binding partners, and by targeting negative regulators of cell cycle progression. Plays a critical role in early lymphocyte development, where it is essential to prevent apoptosis in lymphoid precursors, allowing them to survive in response to IL7 and undergo proper lineage commitment. Has been shown to bind to the promoters of adenovirus major late protein and cyclin D1 and activate transcription. Required for early embryonic development during gastrulation. Represses RB1 transcription; this repression can be blocked by interaction with ZBTB49 isoform 3/ZNF509S1 (PubMed : 25245946).
