The Mkl2 protein also known as Myocardin-like 2 or MRTFB is a transcriptional coactivator that primarily interacts with serum response factor (SRF) to regulate gene expression. It has a molecular weight of approximately 118 kDa. Mkl2 is ubiquitously expressed but shows high levels of expression in heart and skeletal muscles. It predominantly resides in the cytoplasm but translocates to the nucleus upon activation.
Mkl2 influences cytoskeletal dynamics and cellular motility by modulating actin cytoskeleton organization. It forms a complex with SRF to control the transcription of genes involved in these processes. This complex is essential for responding to environmental cues that promote cell differentiation and proliferation.
Mkl2 is integral to the RhoA-MRTFA-SRF signaling pathway which links changes in the actin cytoskeleton to transcriptional responses. Mkl2 works in conjunction with proteins like MKL1 to mediate these pathway interactions. Together they coordinate responses to external stimuli that affect cellular architecture and movement.
Mkl2 has implications in cardiac hypertrophy and cancer progression. Aberrant expression or regulation of Mkl2 can lead to pathological cardiac growth impacting heart function. It also associates with oncogenes in the context of cancer promoting tumor cell invasion and metastasis. Understanding its interactions with proteins like MRTFA in these conditions provides potential therapeutic targets for treatment.
