HepG2 (human liver hepatocellular carcinoma cell line) cells stained for Mkln1 (green) using ab236986 at 1/133 dilution in ICC/IF, followed by Alexa Fluor 488-conjugated goat anti-rabbit IgG (H+L). Counterstained with DAPI (blue).

The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubate with primary antibody at 4°C overnight.

Paraffin-embedded human prostate tissue stained for Mkln1 using ab236986 at 1/400 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum for 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4° overnight. The primary is detected by a biotinylated secondary and visualized using an HRP conjugated SP system.

Mkl1 also known as megakaryoblastic leukemia (translocation) 1 is a protein encoded by the MKL1 gene. The mass of Mkl1 approximately measures around 130 kDa. This protein is expressed in multiple tissues but it exhibits higher expression in the heart and skeletal muscle. Mkl1 localizes mainly in the nucleus where it functions as a coactivator of serum response factor (SRF) playing an important role in actin dynamics and transcriptional regulation.
Biological function summary

Mkl1 coordinates transcriptional programs required for cell differentiation and cytoskeletal organization. As a component of the MRTF-SRF complex Mkl1 regulates genes involved in actin cytoskeleton remodeling. This function impacts cell migration adhesion and morphology influenced by external signals. Additionally it plays a role in the regulation of smooth muscle and cardiac-specific gene expression which highlights its importance in muscle physiology.

Pathways

Mkl1 serves an important function in the RhoA-actin signaling pathway. This pathway controls the organization of the actin cytoskeleton and is connected with several cellular functions including cell shape and motility. Mkl1 interacts closely with SRF as well as other proteins like RhoA GTPase which are vital for transmitting signals that promote actin polymerization and cell movement. This pathway integration contributes to understanding how extracellular cues translate into cytoskeletal rearrangements.

Mkl1 has significant connections to megakaryoblastic leukemia and cardiac hypertrophy. In leukemia a chromosomal translocation involving Mkl1 and another protein MOZ leads to the production of an oncogenic fusion protein that causes dysregulation of normal gene expression contributing to malignancy. Additionally upregulated Mkl1 expression links to cardiac hypertrophy where it facilitates pathological gene expression in heart muscle cells. The role of Mkl1 in these disorders highlights its significance in disease progression and susceptibility.
