Immunocytochemistry/ Immunofluorescence - Anti-MKLP1 antibody [EPR10879] - BSA and Azide free (AB240178)

Immunofluorescence staining of HeLa cells with purified ab174304 at a working dilution of 1 in 400, counter-stained with DAPI. Tubulin was stained with mouse anti-tubulin at a dilution of 1/1000 (ab7291) and Alexa Fluor® 594 goat anti-mouse at a dilution of 1/500 (ab150120) . The secondary antibody was ab150077 Alexa Fluor® 488 goat anti rabbit, used at a dilution of 1 in 500. The cells were fixed in 4% PFA and permeabilized using 0.1% Triton X 100. The negative controls are shown in the bottom middle and right hand panels - for the first negative control, purified ab174304 was used at a dilution of 1/200 followed by an Alexa Fluor® 555 goat anti-mouse antibody at a dilution of 1/500 and for the second negative control mouse primary antibody (ab7291) and anti-rabbit secondary antibody (ab15007) were used.

MKLP1 also known as mitotic kinesin-like protein 1 is a motor protein with a molecular weight of around 95 kDa. This protein plays an important role in cell division by facilitating the movement of chromosomes and the formation of the spindle apparatus during mitosis. MKLP1 is widely expressed in proliferating cells and tissues especially in those undergoing rapid cell cycles such as embryonic tissues and tumors.
Biological function summary

MKLP1 functions as a component of the centralspindlin complex where it interacts with other proteins to regulate cytoskeletal dynamics. It is critical in the formation and resolution of the central spindle during mitosis ensuring proper separation of cells. By coordinating microtubule bundling and motor activities MKLP1 assists in the maintenance of cell polarity and cell shape changes necessary for mitosis completion.

Pathways

MKLP1 is integral to the mitotic and cell cycle pathways. In particular it is important in the mechanics of the spindle assembly checkpoint which ensures chromosomes are correctly segregated before cell division proceeds. MKLP1 interacts with proteins like Aurora B kinase and ECT2 which regulate the dynamics of cytoskeletal reorganization during these critical phases.

MKLP1 has associations with cancers and neurodevelopmental disorders. In tumors overexpression or mutation of MKLP1 often leads to aneuploidy contributing to tumor progression and metastasis. In the context of neurodevelopmental disorders disruptions in MKLP1 function can relate to conditions such as microcephaly due to its role in cell proliferation. Altered interactions between MKLP1 and proteins like BRCA2 in cancer highlight its relevance in both cell division and stability of the genome.
