MLC1 also known as the Megalencephalic Leukoencephalopathy with subcortical Cysts 1 protein is a membrane protein with a molecular mass of approximately 24 kDa. It expresses mainly in the brain especially in astrocytes and oligodendrocytes. MLC1 localizes in the perivascular astrocytic processes which indicates its involvement in maintaining ion homeostasis in the brain. Its primary function revolves around cellular processes related to water and ion transport.
Biological function summary

The MLC1 protein is linked to the regulation of ion channels and is part of a larger protein complex on the astrocyte cell membranes. This complex plays a role in the astrocytic response to osmotic stress contributing to cellular volume regulation. MLC1 influences critical cellular mechanisms through its interactions with other proteins in the complex helping to stabilize ion concentrations and supporting proper brain function under various stress conditions.

Pathways

MLC1 contributes significantly to the osmoregulatory signaling pathways and modulates the astrocyte-neuron communication network. It has interactions with proteins like GlialCAM and ClC-2 which are essential components of the regulatory pathways controlling astrocyte ion homeostasis. These pathways ensure the maintenance of electrochemical gradients important for neuronal signaling and overall brain homeostasis.

MLC1 links directly to megalencephalic leukoencephalopathy with subcortical cysts (MLC) a rare neurodegenerative disorder. Mutations in the MLC1 gene often result in dysfunctional protein which leads to improper ion and water balance in the brain. This condition associates with cognitive and motor deficits. It also connects with the GlialCAM protein which can exacerbate or contribute to the development of MLC when mutated highlighting the importance of the MLC1-GlialCAM interaction in maintaining brain health.
