MMP20 also known as Enamelysin is a member of the matrix metalloproteinase (MMP) family. This enzyme has a molecular mass of approximately 54 kDa. MMP20 is mainly expressed in the dental enamel but researchers have also observed its expression in other tissues like the epithelial and mesenchymal cells of the developing tooth. The enzyme plays a role in the breakdown of extracellular matrix components specifically during the formation of tooth enamel where it processes the nascent enamel matrix proteins.
Biological function summary

MMP20 participates in enamel development and maturation. This enzyme cleaves amelogenin an important protein in the enamel matrix to aid in proper enamel mineralization. MMP20 does not function alone; it exists as part of the complex processes necessary for the tooth structure formation. By regulating protein processing MMP20 ensures the replacement of organic matrix with inorganic mineral important for the hardness of enamel.

Pathways

The involvement of MMP20 is critical in the enamel mineralization pathway and intralays network of amelogenesis. It connects with other proteins like ameloblastin and enamelin which collaborate in enamel formation. MMP20 affects these pathways by enabling the proteolytic processing of enamel-specific proteins supporting the structural changes and mineral deposition essential for hard enamel formation.
