The 'mnt' protein known as Max's Next Tango is a transcriptional repressor with a significant role in gene expression regulation. Mnt has a molecular mass of approximately 34 kDa and belongs to the basic helix-loop-helix (bHLH) family. It forms homodimers or heterodimers with Max another member of the bHLH family. Mnt is expressed in several tissues and cell types including liver brain and muscle tissue. Its presence in diverse tissues indicates a multifaceted role in basic cellular function.
Biological function summary

Mnt functions to regulate cell proliferation and differentiation by acting as a negative regulator. Mnt often forms a complex with Max to repress the transcription of Myc target genes. Myc proteins promote cell cycle progression and growth while Mnt exerts an opposing effect by working as a transcriptional repressor. This delicate balance between Myc activation and Mnt repression is necessary to maintain controlled cell proliferation.

Pathways

Mnt regulates essential cellular processes through the Myc/Max/Mad network and the E2F transcription factor pathway. Within the Myc/Max/Mad network mnt balances out the activity of Myc by forming a competing dimer with Max. In its interaction with the E2F pathway mnt indirectly influences cell cycle progression. These pathways include numerous proteins like Mad1 and Mad3 which also interact with Mnt to modulate transcription.

Mnt has a recognized connection with cancer and neurodegenerative diseases. Abnormal expression or mutations in mnt can disrupt the balance of cell proliferation often leading to tumorigenesis. In neurodegenerative diseases improper regulation of Myc and Mnt affects neurogenesis and neuronal survival. Proteins like p53 often implicated in these diseases show interconnected pathways with Mnt highlighting its potential as a therapeutic target.
