MOCS3 also known as Molybdenum Cofactor Synthesis 3 is an important enzyme involved in the biosynthesis of the molybdenum cofactor. It has a molecular mass of approximately 101 kDa. This enzyme localizes mainly in the cytoplasm and plays a role as a partner in the transfer of sulfur in the form of a thiocarboxylate to L-cysteine. MOCS3 helps catalyze the synthesis of the MPT precursor Z ligand which is important for the formation of active molybdenum cofactor a molecule required for various enzymatic reactions in the body.
Biological function summary

MOCS3 is integral to the tRNA thiolation and molybdenum cofactor biosynthesis processes. It participates in the formation of molybdopterin an organophosphate essential for the activity of the molybdenum enzymes. MOCS3 forms a complex with MOCS2A and MOCS2B collaborating to activate these enzymes. This association highlights its importance in ensuring the synthesis of vital cofactors that impact enzyme function.

Target data

Plays a central role in 2-thiolation of mcm(5)S(2)U at tRNA wobble positions of cytosolic tRNA(Lys), tRNA(Glu) and tRNA(Gln). Also essential during biosynthesis of the molybdenum cofactor. Acts by mediating the C-terminal thiocarboxylation of sulfur carriers URM1 and MOCS2A. Its N-terminus first activates URM1 and MOCS2A as acyl-adenylates (-COAMP), then the persulfide sulfur on the catalytic cysteine is transferred to URM1 and MOCS2A to form thiocarboxylation (-COSH) of their C-terminus. The reaction probably involves hydrogen sulfide that is generated from the persulfide intermediate and that acts as a nucleophile towards URM1 and MOCS2A. Subsequently, a transient disulfide bond is formed. Does not use thiosulfate as sulfur donor; NFS1 acting as a sulfur donor for thiocarboxylation reactions.
