Nuclear matrix protein which forms MORC3-NBs (nuclear bodies) via an ATP-dependent mechanism and plays a role in innate immunity by restricting different viruses through modulation of the IFN response (PubMed:27440897, PubMed:34759314). Mechanistically, possesses a primary antiviral function through a MORC3-regulated element that activates IFNB1, and this function is guarded by a secondary IFN-repressing function (PubMed:34759314). Sumoylated MORC3-NBs associates with PML-NBs and recruits TP53 and SP100, thus regulating TP53 activity (PubMed:17332504, PubMed:20501696). Binds RNA in vitro (PubMed:11927593). Histone methylation reader which binds to non-methylated (H3K4me0), monomethylated (H3K4me1), dimethylated (H3K4me2) and trimethylated (H3K4me3) 'Lys-4' on histone H3 (PubMed:26933034). The order of binding preference is H3K4me3 > H3K4me2 > H3K4me1 > H3K4me0 (PubMed:26933034).(Microbial infection) May be required for influenza A transcription during viral infection (PubMed:26202233).

MORC3 also known as Microrchidia CW-type zinc finger protein 3 is a nuclear protein with a molecular weight of approximately 100 kDa. It functions as part of chromatin remodeling processes and is found within the nucleus of cells. MORC3 exhibits a high level of expression in testes and other tissues supporting its role in transcription regulation and nuclear body formation. It also contains several domains such as ATPase zinc finger and a nuclear localization signal which are critical for its mechanical action within the cell.
MORC3 influences nuclear dynamics and chromatin organization. It plays a central role in the formation of promyelocytic leukemia nuclear bodies (PML-NBs) which are involved in gene regulation DNA repair and apoptosis. MORC3 can interact with proteins within PML-NBs indicating it’s part of a protein complex that maintains genomic stability. This organizational role supports various cellular processes by regulating gene expression patterns in response to stress and damage.
MORC3 integrates into different signaling pathways including DNA damage response and gene repression-specific pathways. It participates in the p53 signaling pathway where its interaction with proteins like p300 influences transcriptional activation of p53 target genes. Moreover MORC3's involvement in these pathways highlights processes such as cell cycle regulation apoptosis and maintenance of genomic integrity under stress conditions.
MORC3 has been associated with autoimmune conditions and cancer. Variants or dysregulation in MORC3 expression can relate to systemic lupus erythematosus where it impacts immune responses through proteins like TRIM21 known for its role in autoimmunity. Additionally changes in MORC3 expression levels have correlations with certain cancer types where aberrant nuclear body formation affects cell proliferation and survival mechanisms.
