Target data

Function

Component of the 60-80S U3 small nucleolar ribonucleoprotein (U3 snoRNP). Required for the early cleavages during pre-18S ribosomal RNA processing (PubMed:12655004). Part of the small subunit (SSU) processome, first precursor of the small eukaryotic ribosomal subunit. During the assembly of the SSU processome in the nucleolus, many ribosome biogenesis factors, an RNA chaperone and ribosomal proteins associate with the nascent pre-rRNA and work in concert to generate RNA folding, modifications, rearrangements and cleavage as well as targeted degradation of pre-ribosomal RNA by the RNA exosome (PubMed:34516797).

Activity summary

MPP10 also known as M-phase phosphoprotein 10 is a component of the nucleolar pre-40S ribosomal subunit. It has a molecular mass of approximately 85 kDa. MPP10 is expressed mainly in the nucleolus of eukaryotic cells where it plays a role in the assembly of ribosomal subunits. This protein acts as a scaffolding structure providing support and regulation necessary for proper ribosome biogenesis. Researchers have identified MPP10 with its involvement in nucleolar processes vital for cell growth and proliferation.

Biological function summary

The involvement of this protein in ribosomal RNA (rRNA) processing is significant. MPP10 is a part of the U3 small nucleolar ribonucleoprotein (snoRNP) complex essential for early cleavages of the pre-rRNA. This function helps initiate the maturation of the 18S rRNA which is a component of the small ribosomal subunit. The presence of MPP10 within this complex indicates its critical role in maintaining efficiency and fidelity during ribosome assembly. Its function impacts cellular protein synthesis influencing cellular growth and division.

Pathways

MPP10 plays an important role in ribosome biogenesis and related molecular pathways. It interacts within the nucleolar processing pathway of pre-18S ribosomal RNA. This pathway important for cell replication involves other proteins such as Fibrillarin and NOP56. MPP10's precise interaction with these proteins supports the maturation of pre-rRNA thereby ensuring the production of functional ribosomal subunits. Disruptions in these pathways can result in imbalances in cellular homeostasis affecting overall cellular function.

Associated diseases and disorders

MPP10 has associations with pathological conditions related to ribosome dysfunction. Abnormal expression or mutations of MPP10 can contribute to disorders like cancer where inappropriate ribosome biogenesis accelerates tumor cell proliferation. Furthermore MPP10's association with other nucleolar proteins such as Dyskerin links it to ribosomopathies a group of disorders characterized by defective ribosome function. Understanding these connections helps researchers target ribosome-related pathways for therapeutic interventions.
