MRAP2 also known as Melanocortin-2 Receptor Accessory Protein 2 is a protein that modulates the activity of melanocortin receptors primarily MC4R. It has a molecular mass of about 16 kDa and is expressed in several regions of the brain including the hypothalamus. MRAP2 plays a role in supporting the function of these receptors by facilitating their trafficking and increasing their functionality on the cell surface.
Biological function summary

MRAP2 interacts with the melanocortin system which involves energy balance and appetite regulation. It forms a complex with melanocortin receptors especially MC4R and to some extent MC3R influencing signaling pathways that control food intake. This system is fundamental in the regulation of metabolism reflecting the importance of MRAP2 in maintaining energy homeostasis in organisms.

Pathways

MRAP2 operates within the melanocortin pathway interacting closely with the leptin signaling pathway which governs energy expenditure and appetite. MRAP2 supports the function of MC4R which in turn relates to the regulation of adenylyl cyclase activity affecting cAMP levels. Additionally MRAP2 has links with genetic conditions involving MC4R that contribute to weight disorders.

Modulator of melanocortin receptor 4 (MC4R), a receptor involved in energy homeostasis. Plays a central role in the control of energy homeostasis and body weight regulation by increasing ligand-sensitivity of MC4R and MC4R-mediated generation of cAMP (By similarity). May also act as a negative regulator of MC2R : competes with MRAP for binding to MC2R and impairs the binding of corticotropin (ACTH) to MC2R. May also regulate activity of other melanocortin receptors (MC1R, MC3R and MC5R); however, additional evidence is required in vivo.
