Supplementary information

MRGPRX4 also known as Mas-related G-protein coupled receptor member X4 is a target involved in several physiological processes. This G-protein coupled receptor (GPCR) has a molecular weight that approximately ranges around 37 kDa. It is part of a larger family of GPCRs and localizes mainly in human tissues predominantly within the brain and skin. MRGPRX4 contributes to signal transduction upon activation by endogenous ligands leading to diverse cellular responses.
Biological function summary

This receptor plays a role in sensory perception and possibly in the detection of noxious stimuli. It interacts within a complex network of receptors involved in signaling pathways related to sensory responses. MRGPRX4 is thought to be significant in modulating physiological responses to external stimuli such as pruritogenic compounds. The receptor likely forms functional complexes with other GPCRs facilitating coordinated signaling events that are essential for adaptive responses.

Pathways

MRGPRX4 is implicated in nociceptive signaling and pruritus pathways. It shares close relationships with other MRGPR family members involved in these pathways. Through these interactions MRGPRX4 influences the transmission of signals that alert the organism to harmful stimuli coordinating with related proteins to mediate perception and reaction. It facilitates the conversion of external chemical stimuli into intracellular actions leading to appropriate organismal responses.

MRGPRX4 shows a connection with chronic itch conditions and neuropathic pain. Abnormalities in the function or expression of MRGPRX4 might contribute to these disorders. The receptor associates with other sensory receptors and ion channels such as TRPV1 within these pathologic contexts. Understanding the role of MRGPRX4 in these conditions provides potential avenues for therapeutic interventions targeting its signaling mechanisms.

Target data

Orphan receptor. Probably involved in the function of nociceptive neurons. May regulate nociceptor function and/or development, including the sensation or modulation of pain. Potently activated by enkephalins (By similarity).
