MRPS16 also known as mitochondrial ribosomal protein S16 is a component of the small subunit of the mitochondrial ribosome. This protein plays an important role in mitochondrial protein synthesis and exhibits a molecular mass of approximately 15 kDa. It is expressed in many tissues where energy production is essential such as the brain muscle and liver. It participates actively in the assembly of the mitochondrial ribosome facilitating the translation of ribosomal RNA into functional proteins important for mitochondrial function.
Biological function summary

Mitochondrial ribosomal protein S16 supports mitochondrial gene expression as part of the mitochondrial ribosomal small subunit. It works in concert with other mitochondrial ribosomal proteins to ensure the efficient translation of mitochondrial DNA-encoded proteins which are integral to oxidative phosphorylation. This protein functions as part of a larger complex the mammalian mitochondrial ribosome which maintains the cellular energy balance by promoting mitochondrial protein synthesis.

Pathways

MRPS16 is an important player in mitochondrial translation and energy production pathways. It contributes to the proper functioning of the oxidative phosphorylation pathway which is critical for ATP generation in cells. MRPS16 functionally interacts with other mitochondrial ribosomal proteins such as MRPS18 in these pathways enabling the full capability of the mitochondrial translation machinery to synthesize proteins necessary for electron transport chain assembly and maintenance.

Mitochondrial dysfunction involving MRPS16 can contribute to conditions such as mitochondrial encephalomyopathy and Leigh syndrome. These disorders often stem from impaired mitochondrial function and energy production where MRPS16 mutation may disrupt core cellular energy processes. The protein's relationship to these diseases highlights possible associations with related mitochondrial proteins like MRPL3 which may share overlapping roles or compensatory mechanisms in mitochondrial ribosome formation and function.
