Supplementary information

MRS2 also known as HPT is a magnesium ion channel primarily found in the mitochondria. The mass of MRS2 is approximately 49 kDa. Expression of MRS2 occurs widely across different tissues indicating its essential role in cellular function. This channel facilitates the transport of magnesium ions ensuring proper mitochondrial function and contributing to cellular energy production.
Biological function summary

MRS2 acts as a critical component in the maintenance of mitochondrial magnesium homeostasis. It operates as part of the mitochondrial membrane complex allowing magnesium ions to enter the organelle. Magnesium ions are vital for numerous enzymatic processes and maintaining mitochondrial stability. Therefore MRS2's function impacts various cellular metabolic processes by supporting the enzymatic machinery within mitochondria.

Pathways

Various mitochondrial processes depend heavily on MRS2 to regulate ionic balance and support energy metabolism. The protein aligns itself with the oxidative phosphorylation pathway where it affects ATP production. Moreover MRS2 interacts with the Letm1 protein reflecting its significance in maintaining mitochondrial health and ionic equilibrium. Together these proteins ensure optimal mitochondrial performance linking MRS2 to important aspects of cellular energy pathways.

Improper function of MRS2 associates with conditions such as hypomagnesemia and familial migraines. MRS2 dysfunction can disrupt magnesium ion homeostasis leading to clinical manifestations. Additionally interactions between MRS2 and the mitochondrial protein Letm1 relate the target to epilepsy as disturbances in these interactions may exacerbate mitochondrial deficiencies linked to the disorder.

Target data

Magnesium transporter that mediates the influx of magnesium into the mitochondrial matrix and regulates magnesium metabolism (PubMed : 11401429, PubMed : 18384665, PubMed : 37543649, PubMed : 37938562, PubMed : 39609651). Also permeable to calcium, sodium and potassium ions (PubMed : 39609651). Required for normal expression of the mitochondrial respiratory complex I subunits (PubMed : 18384665). May play a role in maintaining the inner mitochondrial membrane potential (PubMed : 39609651).
