Immunoprecipitation - Anti-MSH4 antibody [EPR27244-57] (AB313331)
MSH4 was immunoprecipitated from 0.35 mg Mouse testis tissue lysate with ab313331 at 1/30 dilution (2ug in 0.35mg lysates). Western blot was performed on the immunoprecipitate using ab313331 at 1/1000 dilution. VeriBlot for IP secondary antibody(HRP)(ab131366) was used at 1/5000 dilution. Lane 1 : Mouse testis tissue lysate Lane 2 : Mouse testis tissue lysate Lane 3 : Rabbit monoclonal IgG (ab172730) instead of ab313331 in mouse testis tissue lysate Blocking and dilution buffer and concentration : 5% NFDM/TBST. Biological function summary
MSH4 contributes to genome diversity and stability by facilitating the formation and resolution of crossover events during meiosis I. MSH4 forms a heterodimer with MSH5 another MutS homolog which is necessary for recognizing and binding to DNA double-strand breaks. This complex acts as a scaffold for the assembly of other repair proteins required for recombination. The interaction of MSH4-MSH5 complex with important proteins like MLH1 and MLH3 fosters the maturation of crossovers further ensuring genetic diversity and error-free distribution of genetic material.
Pathways
MSH4 plays an important role in meiotic homologous recombination and mismatch repair pathways. Within the homologous recombination pathway MSH4 operates with MSH5 to promote synapsis and crossover formation between homologous chromosomes. This involvement is critical for fertility and genome stability. The protein also interacts with the MLH1 and MLH3 in the repair pathway emphasizing its role in the maintenance and integrity of genetic material during cell division.
