Supplementary information

MSH4 known as MutS Homolog 4 is a protein involved in meiotic recombination and DNA repair. MSH4 is a member of the DNA mismatch repair (MMR) system which is known for its role in maintaining genome stability. The protein has an estimated mass of around 103 kDa. MSH4 is expressed in the testis and ovary where it localizes to the synaptonemal complex during the early stages of meiosis. It assists in the proper alignment and pairing of homologous chromosomes essential for accurate genetic recombination.
Biological function summary

MSH4 contributes to genome diversity and stability by facilitating the formation and resolution of crossover events during meiosis I. MSH4 forms a heterodimer with MSH5 another MutS homolog which is necessary for recognizing and binding to DNA double-strand breaks. This complex acts as a scaffold for the assembly of other repair proteins required for recombination. The interaction of MSH4-MSH5 complex with important proteins like MLH1 and MLH3 fosters the maturation of crossovers further ensuring genetic diversity and error-free distribution of genetic material.

Pathways

MSH4 plays an important role in meiotic homologous recombination and mismatch repair pathways. Within the homologous recombination pathway MSH4 operates with MSH5 to promote synapsis and crossover formation between homologous chromosomes. This involvement is critical for fertility and genome stability. The protein also interacts with the MLH1 and MLH3 in the repair pathway emphasizing its role in the maintenance and integrity of genetic material during cell division.

Defects in the MSH4 function can lead to anomalies in meiotic recombination increasing the risk for infertility and certain cancers. It has been associated with conditions such as azoospermia where dysfunctional MSH4 may result in improper chromosomal segregation. The interaction of MSH4-MSH5 with other proteins like MLH1 also relates to Lynch syndrome characterized by increased cancer propensity due to mismatch repair deficiencies. Therefore understanding MSH4 and its connections with other proteins is essential for researching therapeutic interventions in these conditions.
