Supplementary information

MSRB3 also known as Methionine Sulfoxide Reductase B3 is an enzyme that plays an important role in the reduction of methionine sulfoxide to methionine. This protein is about 25 kilodaltons in mass. MSRB3 is expressed in various tissues with notable presence in the liver kidneys and skin. Its cellular localization often highlights its activity and function within these tissues suggesting its role in maintaining cellular homeostasis.
Biological function summary

Methionine Sulfoxide Reductase B3 protects cells against oxidative stress by repairing oxidized methionine residues in proteins. It serves as an important player in protein repair mechanisms. MSRB3 acts independently and is not known to form parts of larger complexes. This function makes it essential for cellular resilience against oxidative damage and supports the integrity of cellular proteins under stress conditions.

Pathways

MSRB3 functions mainly within the oxidative stress response pathway where it collaborates with methionine sulfoxide reductase A (MSRA) another enzyme with a similar function. Collectively these proteins help restore oxidized methionine residues an important step in maintaining protein structure and function. MSRB3 also influences the methionine cycle which connects to broader processes such as cellular antioxidant defense.

Methionine Sulfoxide Reductase B3 has associations with hearing loss and age-related cataracts. Mutations in the MSRB3 gene can lead to a form of nonsyndromic hearing loss pointing to its significance in auditory function. The connection of MSRB3 with oxidative stress also implicates it in cataract formation where proteins like alpha-crystallins interact within the lens to maintain transparency and function. Understanding these connections provides insight into therapeutic targets for these conditions.
