The protein kinases MST4 MST3 and STK25 are part of the Ste20 kinase family. MST4 and MST3 are often referred to as serine/threonine-protein kinases 26 and 24 respectively while STK25 is known as Ste20-like kinase. These proteins share a common modular architecture consisting of a kinase domain and various regulatory regions. The molecular masses are approximately 50 kDa for MST4 and MST3 and around 54 kDa for STK25. You can find these proteins expressed in multiple tissues including brain testis and spleen.
Biological function summary

MST4 MST3 and STK25 regulate cell growth proliferation and apoptosis. These proteins interact with various cellular components to form signaling complexes that influence cellular responses. MST3 and MST4 can activate apoptosis signal-regulating kinase 1 (ASK1) which leads to the apoptosis pathway. STK25 plays a role in the Golgi apparatus' functionality affecting cellular trafficking and signal transduction.

Pathways

MST4 MST3 and STK25 are involved in critical signaling pathways like the Hippo pathway and MAPK signaling. Within these pathways MST4 and MST3 together associate with MOB1 and LATS1/2 enhancing their functions. STK25 interacts with STRAD and LKB1 contributing to cell polarity regulation and cellular response to external stimuli.

Altered expression and mutations in MST4 MST3 and STK25 associate with cancer and neurological disorders. Cancer progression links strongly with these kinases' dysregulation due to their role in apoptosis and cell proliferation where MST4 activates CST complex involved in these diseases. Neurodegenerative disorders involve STK25 connected via its regulatory influence on cellular structures and its interactions with Huntington protein influencing disease pathophysiology.

Targets to the Golgi apparatus where it appears to regulate protein transport events, cell adhesion, and polarity complexes important for cell migration. Part of the striatin-interacting phosphatase and kinase (STRIPAK) complexes. STRIPAK complexes have critical roles in protein (de)phosphorylation and are regulators of multiple signaling pathways including Hippo, MAPK, nuclear receptor and cytoskeleton remodeling.
