Supplementary information

MTIF3 also known as mitochondrial translational initiation factor 3 (IF-3mt) weighs approximately 29 kDa. This protein facilitates the initiation of translation within mitochondria by ensuring the correct assembly of ribosomes. MTIF3 expression occurs in tissues with high energy demands such as heart and skeletal muscles due to its role in mitochondrial function. Notably the protein aids in disassociation of the ribosomal subunits enabling the correct positioning of mRNA on the ribosome.
Biological function summary

Mitochondrial translation regulation relies on MTIF3's interaction with other translation factors. MTIF3 as part of the mitochondrial translation initiation complex interacts with mitochondrial ribosomes to prepare them for protein synthesis. This protein regulates the precision of translation initiation ensuring that the translation machinery only begins under appropriate conditions. MTIF3 binds to the small ribosomal subunit playing a guiding role at the beginning of mitochondrial translation.

Pathways

MTIF3 integrates into processes involving mitochondrial protein synthesis and energy production. MTIF3's function significantly affects key pathways like oxidative phosphorylation as it ensures the efficient production of mitochondrial proteins needed for this process. Its association with other factors such as MTIF2 another initiation factor further characterizes its role in this pathway. Together these proteins are critical for the production of proteins that compose the electron transport chain necessary for ATP generation.

MTIF3 has implications for conditions linked to mitochondrial dysfunction. Researchers have identified its role in mitochondrial cardiomyopathy where defects in MTIF3 may lead to impaired cardiac energy metabolism. Potential connections also exist between MTIF3 and certain mitochondrial encephalomyopathies where its impact on mitochondrial translation affects neurological function. Disruptions in MTIF3 levels may alter the expression of other mitochondrial proteins exacerbating such conditions.

Target data

IF-3 binds to the 28S ribosomal subunit and shifts the equilibrium between 55S ribosomes and their 39S and 28S subunits in favor of the free subunits, thus enhancing the availability of 28S subunits on which protein synthesis initiation begins.
