Supplementary information

Munc13-3 also known as Unc-13 homolog C (UNC13C) is a protein involved in synaptic vesicle priming necessary for neurotransmitter release. It weighs approximately 230 kDa. Munc13-3 is expressed mainly in the brain specifically in regions such as the cerebellum. This protein plays a role in the efficiency and timing of synaptic transmission which is critical for normal neuronal communication.
Biological function summary

Munc13-3 acts in the presynaptic region where it functions as part of the synaptic vesicle release machinery. It forms a complex with other proteins like syntaxin Munc18 and RIM proteins to facilitate neurotransmitter release. By interacting with these molecules it helps prepare vesicles for fusion with the plasma membrane modulating synaptic strength and plasticity.

Pathways

Proteins involved in synaptic transmission and plasticity often associate with Munc13-3. One important pathway is the synaptic vesicle cycle where Munc13-3 works closely with synaptotagmin and SNARE proteins to regulate vesicle docking and fusion. Another relevant pathway is the calcium signaling pathway which triggers vesicle release through Munc13-3's interactions with other calcium-sensing proteins ensuring precise neurotransmitter release in response to calcium influx.

Links between Munc13-3 and neurological conditions like autism spectrum disorders and epilepsy are evident. Munc13-3 influences synaptic function and alterations in its expression or function can disrupt normal synaptic activity. Related proteins are Munc13-1 and STXBP1 which interact with Munc13-3 in these conditions suggesting possible therapeutic targets for modulating synaptic communication issues seen in these disorders.
