Munc18-2 also known as Syntaxin-binding protein 2 is a 65 kDa protein that plays a role in vesicle transport machinery. It interacts with syntaxins to regulate the docking and fusion of vesicles with their target membranes. Munc18-2 is mainly expressed in hematopoietic cells where it facilitates regulated secretion.
Biological function summary

This protein facilitates the formation of secretory vesicle complexes. Munc18-2 is a part of the SNARE complex working synergistically with other components to ensure efficient vesicular transport and release. In immune cells this supports processes such as the exocytosis of lytic granules and cytokine release essential for immune response functions.

Pathways

Munc18-2 plays a role in the exocytosis and vesicular fusion pathways. It is closely associated with the SNARE proteins like VAMPs (vesicle-associated membrane proteins) and SNAPs (synaptosomal-associated proteins). In the immune system it is part of the machinery that mediates the transport and release pathways of granules and other vesicular contents.

Munc18-2 dysfunction is linked to familial hemophagocytic lymphohistiocytosis (FHL). This condition involves defective immune response due to hindered vesicular secretion leading to uncontrolled inflammation. Munc18-2 mutations affect the interaction with syntaxin 11 a SNARE protein which disturbs immune cell function contributing to the pathogenesis of this disorder.
