Activity summary

Myf5 also known as Myogenic Factor 5 is a member of the myogenic regulatory factor (MRF) family. It plays a critical role in muscle differentiation. Myf5 has a molecular mass of approximately 29 kDa. It is expressed predominantly in skeletal muscle tissues. The protein is observed early in muscle development marking the beginning stages where muscle precursor cells commit to becoming muscle cells.

Biological function summary

Myf5 functions as a transcription factor that initiates muscle differentiation. It acts as part of a larger complex interacting with other MRFs such as MyoD and MYF6. These factors together regulate the expression of genes necessary for myoblasts to develop into mature muscle fibers. Myf5 directly binds to the promoter regions of target genes initiating transcription that supports muscle cell lineage progression.

Pathways

The myogenesis pathway is heavily influenced by Myf5 activity. It works in tandem with MyoD in the early specification of myoblasts and their proliferation. Secondary to this Myf5 forms a feedback loop with proteins like MEF2 and Pax7 which reinforces muscle identity. Myf5 expression helps maintain the balance between myoblast proliferation and differentiation ensuring effective muscle regeneration and repair.

Associated diseases and disorders

Muscular dystrophy exemplifies a condition closely associated with Myf5. The protein's expression levels and functionality often correlate with the disease affecting muscle development and integrity. Satellite cells which express Myf5 become important in muscle repair and dysfunctional Myf5 can contribute to disease progression. Another disorder influenced by Myf5 is rhabdomyosarcoma where aberrant expression disrupts normal myogenic regulatory pathways alongside MyoD dysregulation potentially leading to tumorigenesis.
