Mechanically MYO10 also known as Myosin-X is an unconventional myosin motor protein with a molecular mass of approximately 237 kDa. This protein has a significant presence in specialized cellular structures such as filopodia and neuron growth cones. It functions as a motor that transports cellular cargo along actin filaments playing an important role in cellular movement and organization. Expression of MYO10 occurs in various tissues but it is notably abundant in the brain and testis.
Biological function summary

MYO10 facilitates several cellular processes by mediating actin-based transport and cellular structure dynamics. It forms part of a complex network interacting with actin filaments influencing the dynamics of cell protrusions such as filopodia. These processes are essential for cell migration development and intracellular transport where MYO10 acts as an anchor enabling cells to respond to external stimuli and migrate accordingly.

MYO10 interacts with various molecular networks influencing critical signaling cascades. It plays a significant role in the Wnt signaling pathway by interacting with Dishevelled (Dvl) proteins impacting cell polarization and movement. Additionally MYO10 is involved in integrin signaling facilitating the formation of adhesive contacts necessary for cell movement linking proteins such as talin and integrins into the pathway for integrative structural and signal modulation.

MYO10 has connections with certain pathologies particularly cancer and neurological disorders. Target data

Myosins are actin-based motor molecules with ATPase activity. Unconventional myosins serve in intracellular movements. MYO10 binds to actin filaments and actin bundles and functions as a plus end-directed motor. Moves with higher velocity and takes larger steps on actin bundles than on single actin filaments (PubMed : 27580874). The tail domain binds to membranous compartments containing phosphatidylinositol 3,4,5-trisphosphate or integrins, and mediates cargo transport along actin filaments. Regulates cell shape, cell spreading and cell adhesion. Stimulates the formation and elongation of filopodia. In hippocampal neurons it induces the formation of dendritic filopodia by trafficking the actin-remodeling protein VASP to the tips of filopodia, where it promotes actin elongation. Plays a role in formation of the podosome belt in osteoclasts.. Isoform Headless. Functions as a dominant-negative regulator of isoform 1, suppressing its filopodia-inducing and axon outgrowth-promoting activities. In hippocampal neurons, it increases VASP retention in spine heads to induce spine formation and spine head expansion (By similarity).
