NABC1 also known as New ABC Transporter 1 is an ATP-binding cassette transporter protein with a molecular mass of around 160 kDa. Mechanically NABC1 functions as a transporter utilizing the energy from ATP hydrolysis to move lipids across cellular membranes. This protein is widely expressed in many tissues particularly in liver kidneys and intestines. Its transport activity plays a central role in maintaining cellular lipid homeostasis.
Biological function summary

NABC1 influences lipid metabolism by mediating the efflux of cholesterol and phospholipids from cells to lipid-poor apolipoproteins. It is not part of a larger protein complex but interacts dynamically with extracellular and membrane-associated proteins to facilitate lipid transport. These interactions are important for preventing lipid accumulation within cells which is essential for cellular health and normal functioning.

Pathways

NABC1 participates in the reverse cholesterol transport pathway and is integral to the regulation of cholesterol levels. It works closely with proteins such as apolipoprotein A-I to facilitate cholesterol removal from peripheral tissues thereby contributing to the overall cholesterol homeostasis. This pathway helps to prevent the buildup of excess cholesterol and protects against atherosclerosis.

Abnormal NABC1 activity links to conditions like Tangier disease and familial HDL deficiency where lipid transport becomes impaired. Mutations in NABC1 lead to reduced or absent transporter activity resulting in very low levels of HDL cholesterol in the blood. This defect causes cholesterol to accumulate in tissues causing complications associated with these disorders. NABC1's role in these conditions highlights its interaction with other proteins involved in lipid metabolism and cholesterol transport.
