Detection of Human NAGK by Immunoprecipitation and Western Blot. Whole cell lysate from HeLa at 1 mg for IP, 20% of IP loaded. ab95182 was used for IP at 3 mcg/mg lysate and 1ug/ml for wb (lane 2) NAGK was also immunoprecipitated by another rabbit antibody (lane 1) and control IgG (lane 3). Detection : Chemiluminescence with exposure times of 30 seconds.

N-Acetylglucosamine Kinase (NAGK) also known as GlcNAc kinase catalyzes the phosphorylation of N-acetylglucosamine to N-acetylglucosamine-6-phosphate. It has a molecular mass of approximately 37 kDa. NAGK expresses in various tissues prominently within the liver and pancreas indicating its involvement in metabolic functions related to these organs.
Biological function summary

NAGK plays a significant role in amino sugar and nucleotide sugar metabolism. It does not form part of a large protein complex but interacts with other metabolic enzymes to carry out its function. By converting N-acetylglucosamine to its phosphorylated form NAGK helps to maintain cellular energy balance and provides substrates necessary for glycosylation processes.

Converts endogenous N-acetylglucosamine (GlcNAc), a major component of complex carbohydrates, from lysosomal degradation or nutritional sources into GlcNAc 6-phosphate (PubMed : 22692205). Involved in the N-glycolylneuraminic acid (Neu5Gc) degradation pathway : although human is not able to catalyze formation of Neu5Gc due to the inactive CMAHP enzyme, Neu5Gc is present in food and must be degraded (PubMed : 22692205). Also has N-acetylmannosamine (ManNAc) kinase activity (By similarity). Also involved in innate immunity by promoting detection of bacterial peptidoglycan by NOD2 : acts by catalyzing phosphorylation of muramyl dipeptide (MDP), a fragment of bacterial peptidoglycan, to generate 6-O-phospho-muramyl dipeptide, which acts as a direct ligand for NOD2 (PubMed : 36002575).
