Supplementary information

NALP2 also known as NLRP2 or NOD-like receptor protein 2 is a component of the NLR family. The protein has a molecular mass of approximately 124 kDa. NALP2 is expressed in various tissues including those of the immune system and certain epithelial cells. It serves as a sensor for pathogen-associated and damage-associated molecular patterns which enables the initiation of immune responses. NALP2 contains domains such as a pyrin domain (PYD) a NACHT domain and leucine-rich repeats that enable it to interact with other proteins in the innate immune system.
Biological function summary

The function of the NALP2 protein involves modulating immune responses and inflammatory processes. NALP2 associates with other proteins to form multiprotein complexes known as inflammasomes. These complexes play a role in the activation of pro-inflammatory cytokines like IL-1β and IL-18. This protein can influence cell-signaling pathways that determine responses to cellular stress or foreign pathogens.

Pathways

The NALP2 protein takes part in the inflammasome signaling and NF-kB activation pathways. Through these pathways it helps in bridging innate immunity and inflammation. Its activity can impact the function of other related proteins such as NLRP3 and ASC which also play important roles in the formation and activation of inflammasomes. The function of NALP2 in these pathways influences the broader regulatory network of immune responses.

The NALP2 protein has a connection with inflammatory diseases and autoimmune disorders. Altered function or expression of NALP2 can contribute to these conditions due to improper regulation of inflammation. For example dysregulation of NALP2 may influence disorders like rheumatoid arthritis. NALP2's interaction with cytokines like IL-1β links it to the pathogenesis of inflammatory diseases and highlights potential therapeutic targets involving inflammasome-related pathways.

Target data

Suppresses TNF- and CD40-induced NFKB1 activity at the level of the IKK complex, by inhibiting NFKBIA degradation induced by TNF. When associated with PYCARD, activates CASP1, leading to the secretion of mature pro-inflammatory cytokine IL1B. May be a component of the inflammasome, a protein complex which also includes PYCARD, CARD8 and CASP1 and whose function would be the activation of pro-inflammatory caspases.
