NALP4 also known as NLRP4 is a member of the NOD-like receptor (NLR) family. This protein has a mass of approximately 116 kDa. NALP4 is present mainly in immune cells such as macrophages and dendritic cells. It is located within the cytoplasm where it participates in the recognition of specific cellular ligands and forms a multi-protein complex known as the inflammasome.
Biological function summary

NLRP4 plays a role in innate immune response regulation. Part of the inflammasome complex it senses intracellular danger signals and plays a role in inflammatory processes. NLRP4 involves the recruitment of other components such as ASC (apoptosis-associated speck-like protein) and pro-caspase-1 leading to the activation of caspase-1 and the maturation of pro-inflammatory cytokines like IL-1β and IL-18.

Pathways

NLRP4 contributes to the regulation of the inflammatory signaling cascade a subset of the broader immune response pathway. NLRP4 connects with proteins such as NLRP3 and NLRC4 within these pathways highlighting its role in modulating cytokine release and inflammatory responses. By integrating signals NLRP4 influences both protective and potentially harmful inflammation maintaining tissue homeostasis.

NLRP4 has associations with autoimmune conditions and chronic inflammatory diseases. It is linked to diseases such as systemic lupus erythematosus and Crohn’s disease where dysregulation of NLRP4 activities contributes to pathogenesis. In these cases interactions between NLRP4 and proteins like IL-1β or ASC may exacerbate inflammatory processes offering potential targets for therapeutic intervention.

May be involved in inflammation and recognition of cytosolic pathogen-associated molecular patterns (PAMPs) not intercepted by membrane-bound receptors. Acts as a negative regulator of the type I interferon signaling pathway by serving as an adapter to promote DTX4-mediated ubiquitination of activated TBK1, and its subsequent degradation. Suppresses NF-kappaB induction by the cytokines TNFA and IL1B, suggesting that it operates at a point of convergence in these two cytokine signaling pathways.
