The NANOS2 protein also known simply as NANOS 2 is a member of the NANOS family of proteins and plays an integral role in the regulation of germ cell development. It has a molecular mass of approximately 17 kDa. This protein is primarily expressed in the male germline specifically in spermatogonial stem cells within the testes. Its expression is tightly regulated and is essential for the proper maintenance and differentiation of these stem cells contributing significantly to male fertility.
Biological function summary

NANOS2 acts as a post-transcriptional regulator and it is part of a ribonucleoprotein complex. It helps maintain germ cell pluripotency and influences the cell cycle. Functioning mainly through the binding to the 3' untranslated region (3' UTR) of target mRNAs NANOS2 controls the translation and stability of specific mRNAs that are important for germ cell maintenance and differentiation. This control is essential for preventing premature meiosis or differentiation in male germ cells.

Pathways

The involvement of NANOS2 is critical in both the PI3K/AKT signaling pathway and the TGF-beta signaling pathway. In the PI3K/AKT pathway NANOS2 interacts with proteins such as PI3K and AKT to regulate cell survival and proliferation specifically in germ cells. In the TGF-beta pathway NANOS2 works alongside Smad proteins to modulate responses essential for cell cycle regulation ensuring the proper timing of germ cell differentiation.

Disruptions in NANOS2 expression or function are closely related to infertility particularly affecting spermatogenesis. Studies have shown that NANOS2 abnormalities can lead to male infertility due to inadequate spermatogonial stem cell maintenance and differentiation. Additionally NANOS2 has been linked to testicular germ cell tumors with its expression patterns altered in cancerous cells compared to normal tissue. Such pathological conditions often see interactions with oncogenic proteins like PLZF highlighting the importance of NANOS2 in both normal and disease states of germ cell development.

Plays a key role in the sexual differentiation of germ cells by promoting the male fate but suppressing the female fate. Represses the female fate pathways by suppressing meiosis, which in turn results in the promotion of the male fate. Maintains the suppression of meiosis by preventing STRA8 expression, which is required for premeiotic DNA replication, after CYP26B1 is decreased. Regulates the localization of the CCR4-NOT deadenylation complex to P-bodies and plays a role in recruiting the complex to trigger the degradation of mRNAs involved in meiosis. Required for the maintenance of the spermatogonial stem cell population. Not essential for the assembly of P-bodies but is required for the maintenance of their normal state (By similarity).
