The protein NAS6 also known as p28 and gankyrin is a regulatory component with an approximate mass of 28 kDa. NAS6 has significant expression in the human liver as well as detectable levels in several other tissues such as the brain and heart. Mechanically NAS6 interacts with the proteasome aiding in the regulation of protein degradation. The interaction with the proteasome involves NAS6 binding to specific subunits and helping in substrate recognition.
Biological function summary

NAS6 plays an important role in cellular protein homeostasis by associating with the 26S proteasome complex. This association facilitates the targeting and degradation of ubiquitinated proteins which is critical for maintaining cellular function. Through its activity NAS6 contributes to the removal of damaged or misfolded proteins and regulation of the cell cycle. This regulation includes controlling the abundance of cyclins and other proteins that are pivotal in cell division processes.

Pathways

NAS6 integrates into the ubiquitin-proteasome pathway a highly conserved pathway responsible for protein turnover in the cell. The pathway involves proteins like ubiquitin which tags proteins for degradation and the 26S proteasome where NAS6 plays its regulatory role. NAS6 also interacts with other proteins such as MDM2 in the p53 pathway linking its function to cellular stress responses and apoptosis regulation.

NAS6 has implications in oncogenesis particularly in liver cancer and colorectal cancer. Overexpression of NAS6 links to uncontrolled cell proliferation mostly due to its effect on protein degradation pathways. Additionally NAS6 interacts with the retinoblastoma protein (Rb) influencing pathways associated with tumorigenesis. Alterations in NAS6 levels or function can disrupt normal cellular regulation highlighting its relevance in cancer research and therapy development.
