Supplementary information

The NBAS protein also known as Neuroblastoma Amplified Sequence functions as an important player in intracellular trafficking and protein stability. It weighs approximately 237 kDa and is broadly expressed across various tissues with noticeable presence in the liver and brain. The NBAS protein forms part of the syntaxin-binding protein 5 (STXBP5) complex aligning itself with other cytoplasmic and membrane-associated proteins. Through its interaction with these cellular structures the NBAS protein facilitates proper protein transportation within the cells.
Biological function summary

The NBAS protein contributes to maintaining homeostasis by participating in endoplasmic reticulum-to-Golgi apparatus transport. This protein joins the syntaxin 18 complex an assembly of proteins that forms part of the broader secretion machinery in cells. Through this involvement NBAS ensures effective protein sorting and delivery which is critical for cell function and communication. Disruption in NBAS activity might lead to accumulation of unfolded proteins affecting overall cellular health.

Pathways

The NBAS protein plays a significant role in the vesicular transport and secretory pathways. It functions alongside proteins such as STX18 and presumed interactions with other members of the SNARE complex facilitating the fusion of vesicles with target membranes. By participating in these pathways NBAS ensures the smooth transition of proteins and contributes indirectly to processes like secretion and intracellular signaling.

NBAS mutations have been associated with Short Stature Onychodysplasia Facial Dysmorphism and Hypotrichosis (SOFT) syndrome as well as liver dysfunction. SOFT syndrome symptoms indicate the protein's involvement in bone growth and hair follicle development while liver dysfunction suggests its necessity in hepatic protein trafficking. In both cases the NBAS shares connections with proteins in similar pathways such as components of the syntaxin and SNARE family indicating a broader impact on cellular and systemic health when NBAS function is compromised.

Target data

Involved in Golgi-to-endoplasmic reticulum (ER) retrograde transport; the function is proposed to depend on its association in the NRZ complex which is believed to play a role in SNARE assembly at the ER (PubMed : 19369418). Required for normal embryonic development (By similarity). May play a role in the nonsense-mediated decay pathway of mRNAs containing premature stop codons (By similarity).
