NBL1 also known as DAN domain family member 1 is a protein with a molecular weight of approximately 20 kDa. It is found in various tissues but shows higher expression levels in the nervous tissue and skeletal muscle. Mechanically NBL1 acts as a modulator in cell signaling pathways by binding to specific growth factors such as BMPs (Bone Morphogenetic Proteins) inhibiting their functions and affecting cell differentiation and growth.
Biological function summary

NBL1 plays an inhibitory role in bone morphogenesis by interacting with BMPs which are part of the TGF-beta superfamily. NBL1 can form complexes with these growth factors thereby regulating processes related to bone and cartilage development. This protein is essential for maintaining cellular processes involved in tissue homeostasis and suppressing unchecked cellular proliferation.

Pathways

NBL1 involves itself in the BMP signaling pathway where it directly impacts the regulation of bone and cartilage development. It is associated with other proteins like Noggin and Chordin both of which also modulate BMP activity highlighting its importance in balancing cell growth and differentiation signals. The involvement of NBL1 in the TGF-beta/BMP pathway is critical for embryonic development and the regulation of body patterning.

NBL1 has been linked to conditions such as osteoarthritis and certain cancer types.
