What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Species reactivity
Rat: We have preliminary internal testing data to indicate this antibody may not react with this species.
Please contact us for more information.

Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Supplementary information

The protein NCOA62 also known as SNW1 or Ski-interacting protein functions as a transcriptional coactivator in the cell nucleus. This protein with a mass of approximately 60 kDa is involved in various transcriptional activities. You will find it expressed in many tissue types but especially in the highly proliferative ones like the liver brain and reproductive tissues. The protein associates with other nuclear receptors and transcription factors to modulate gene expression.
Biological function summary

NCOA62/SNW1 interacts with several transcription complexes playing an important role in modulating transcriptional signals. It acts as a scaffolding molecule facilitating the assembly and function of specific transcriptional complexes. It binds with various partners including the Smad and p53 proteins partaking in cell cycle regulation and DNA repair processes. Its role in transcription complex suggests its involvement in essential cellular functions.

Pathways

NCOA62/SNW1 plays a role in the TGF-beta signaling pathway and the p53 pathway. These pathways are critical for regulating cell growth differentiation and apoptosis. Within the TGF-beta pathway the protein aids in transmitting signals from the cell membrane to the nucleus by interacting with Smad proteins. It also influences the p53 pathway by modulating p53 activity an important protein recognized for its role in maintaining genomic integrity and responding to cellular stress.

NCOA62/SNW1 displays significant involvement in several medical conditions including cancer and neurodegenerative diseases. In cancer it influences the cell cycle and apoptosis by interacting with Smad and p53 potentially leading to uncontrolled cell growth when dysregulated. Regarding neurodegenerative disorders alterations in NCOA62/SNW1 expression or function could contribute to neuronal cell death by disrupting normal cell signaling mechanisms connected possibly through interaction with p53 well-known for its involvement in cell death and survival pathways.

Target data

Involved in pre-mRNA splicing as component of the spliceosome (PubMed : 11991638, PubMed : 28076346, PubMed : 28502770). As a component of the minor spliceosome, involved in the splicing of U12-type introns in pre-mRNAs (Probable). Required for the specific splicing of CDKN1A pre-mRNA; the function probably involves the recruitment of U2AF2 to the mRNA. May recruit PPIL1 to the spliceosome. May be involved in cyclin-D1/CCND1 mRNA stability through the SNARP complex which associates with both the 3'end of the CCND1 gene and its mRNA. Involved in transcriptional regulation. Modulates TGF-beta-mediated transcription via association with SMAD proteins, MYOD1-mediated transcription via association with PABPN1, RB1-mediated transcriptional repression, and retinoid-X receptor (RXR)- and vitamin D receptor (VDR)-dependent gene transcription in a cell line-specific manner probably involving coactivators NCOA1 and GRIP1. Is involved in NOTCH1-mediated transcriptional activation. Binds to multimerized forms of Notch intracellular domain (NICD) and is proposed to recruit transcriptional coactivators such as MAML1 to form an intermediate preactivation complex which associates with DNA-bound CBF-1/RBPJ to form a transcriptional activation complex by releasing SNW1 and redundant NOTCH1 NICD.. (Microbial infection) Is recruited by HIV-1 Tat to Tat : P-TEFb : TAR RNA complexes and is involved in Tat transcription by recruitment of MYC, MEN1 and TRRAP to the HIV promoter.. (Microbial infection) Proposed to be involved in transcriptional activation by EBV EBNA2 of CBF-1/RBPJ-repressed promoters.
