Supplementary information

NECAB2 also known as N-terminal EF-hand calcium-binding protein 2 binds calcium ions through EF-hand motifs. This protein belongs to a family of EF-hand containing proteins involved in calcium signaling. It has a molecular weight around 45 kDa. NECAB2 is expressed mainly in the nervous system particularly in the brain. Studies show its expression in neurons suggesting a role in synaptic function.
Biological function summary

NECAB2 is involved in calcium-dependent signaling pathways. It associates with other proteins to form complexes impacting cellular processes like neurotransmitter release. NECAB2's role suggests it helps modulate synaptic activity and plasticity due to its involvement in regulating intracellular calcium levels. Its interactions in synaptic regions underline its importance in neural communication.

Pathways

NECAB2 participates in calcium signaling and synaptic signaling pathways. In calcium signaling NECAB2 couples with other proteins like calmodulin affecting intracellular calcium concentrations. In synaptic signaling its interaction with proteins like synaptotagmins impacts neurotransmitter release and synapse maintenance. These pathways highlight NECAB2's role in maintaining neural communication and function.

NECAB2 links to neurological conditions such as autism spectrum disorders and schizophrenia. Studies show altered NECAB2 expression in these conditions possibly disrupting normal synaptic function. Its interactions with proteins like neurexins and neuroligins which are implicated in synapse formation and function might contribute to the pathophysiology of these disorders. Understanding NECAB2 in this context could offer insights into therapeutic targets for related disorders.

Target data

May act as a signaling scaffold protein that senses intracellular calcium. Can modulate ligand-induced internalization of ADORA2A and coupling efficiency of mGluR5/GRM5; for both receptors may regulate signaling activity such as promoting MAPK1/3 (ERK1/2) activation.
