Supplementary information

NEGR1 also known as Neuronal Growth Regulator 1 is a protein that plays an essential role in cellular adhesion and neurodevelopment. It carries a molecular mass of approximately 29 kDa. This protein shows expression mainly in the human brain particularly in regions such as the cortex and cerebellum. Studies have also detected NEGR1 in peripheral tissues to a lesser extent indicating a possible broader range of functions.
Biological function summary

NEGR1 contributes to neuronal growth and connectivity. It forms part of the immunoglobulin superfamily which includes proteins involved in the immune response and cell adhesion processes. NEGR1 interacts with cellular membranes through a glycosylphosphatidylinositol anchor facilitating cell-cell communication and signaling necessary for neural network formation. This interaction allows NEGR1 to be a part of synaptic development and plasticity affecting learning and memory processes.

Pathways

NEGR1 plays a part in neuronal growth pathways and synaptic signaling. These pathways are important for the development and maintenance of neuronal circuits. NEGR1 has connections to the MAPK pathway which oversees cell differentiation and proliferation. It interacts with other synaptic proteins like Neurexins and Neuroligins which are important for synaptic formation and stability underlining its integrative role in neural development.

NEGR1 has links to neurodevelopmental disorders and metabolic conditions. Research has identified associations between NEGR1 variants and autism spectrum disorders suggesting its impact on neural circuit wiring might influence these developmental conditions. Additionally NEGR1 appears related to obesity where it interacts with proteins like Leptin which regulates energy balance and body weight. These associations underline NEGR1's relevance in both neural and metabolic health.
