Supplementary information

NEK8 also known as NIMA-related kinase 8 is a serine/threonine kinase. It measures approximately 73 kDa. This kinase plays a role in cell cycle regulation and is predominantly expressed in the kidney liver and pancreas tissues. NEK8 localizes to centrosomes and cilia which suggests its involvement in the structure and function of these organelles.
Biological function summary

NEK8 functions in the regulation of cilia length and stability. It integrates within a signaling complex at the ciliary transition zone important for ciliary function. NEK8 interacts with other components such as inversin and polycystin-2 essential for maintaining normal ciliary processes and signal transduction. Its role in ciliary dynamics suggests its importance to the proper cellular communication and organ development.

Pathways

NEK8 participates in the cilia-related signaling pathways and cell cycle regulation. It influences the Hedgehog signaling pathway which is vital for various developmental processes. NEK8 also interacts with proteins like Gli family transcription factors within this pathway. Another pathway involving NEK8 is the DNA damage response where it collaborates with proteins like ATR to maintain genomic stability.

NEK8 is linked to nephronophthisis and polycystic kidney disease. Mutations within NEK8 impair normal ciliary functions leading to ciliopathies that affect kidney development and function. PKD1 and PKD2 proteins associated with polycystic kidney disease have correlated functional interactions with NEK8 highlighting its significance in maintaining renal health.

Target data

Required for renal tubular integrity. May regulate local cytoskeletal structure in kidney tubule epithelial cells. May regulate ciliary biogenesis through targeting of proteins to the cilia (PubMed : 37598857). Plays a role in organogenesis, and is involved in the regulation of the Hippo signaling pathway (PubMed : 26967905).
