The protein called Neuron-Derived Neurotrophic Factor (NENF) also known as BEND5 plays important roles in cellular processes. This protein weighs approximately 18 kDa. NENF is predominantly found in the central nervous system including the brain and spinal cord where it influences neural function. The protein is secreted and operates primarily in extracellular spaces where it interacts with receptors to execute its functions.
Biological function summary

The protein NENF influences cell survival and differentiation. It acts as a neurotrophic factor contributing to the development and maintenance of neurons. Although not part of a larger complex NENF interacts directly with receptors on target cells to initiate signaling cascades. This modulating effect on neural cells plays a substantial role in maintaining cellular mechanisms such as synaptic plasticity and neural circuit formation.

Pathways

NENF actively engages in the neurotrophic signaling pathway which is essential for proper nervous system function. Acts as a neurotrophic factor in postnatal mature neurons enhancing neuronal survival (PubMed : 31536960). Promotes cell proliferation and neurogenesis in undifferentiated neural progenitor cells at the embryonic stage and inhibits differentiation of astrocytes (By similarity). Its neurotrophic activity is exerted via MAPK1/ERK2, MAPK3/ERK1 and AKT1/AKT pathways (By similarity). Neurotrophic activity is enhanced by binding to heme (By similarity).
