Dot blot analysis of Nephrin (pY1217) phospho peptide (Lane 1) and Nephrin non-phospho peptide (Lane 2) labelling Nephrin (pY1217) with ab80298 at a dilution of 1/1000. A Goat Anti-Rabbit IgG, (H+L), Peroxidase conjugated (ab97051) was used as the secondary antibody at a dilution of 1/20,000. Blocking buffer : 5% NFDM/TBST. Dilution buffer : 5% NFDM /TBST.
This data was developed using the same antibody clone in a different buffer formulation containing PBS, BSA, glycerol, and sodium azide (ab80298).

Nephrin is essential for the regulation of glomerular permeability and podocyte foot process architecture. It serves as a significant component of the slit diaphragm complex working alongside proteins like podocin and CD2AP. This protein complex helps regulate the integrity and selectivity of the glomerular filtration barrier ensuring that blood plasma contents are selectively filtered while preventing the loss of large proteins into urine.
Pathways
Nephrin is integral to the PI3K/Akt signaling pathway influencing cell survival and cytoskeletal organization in podocytes. Nephrin's interaction within this pathway involves related proteins such as GAPDH and PTPRO which impact cellular functions critical to maintaining kidney structure and function.
