Netrins control guidance of CNS commissural axons and peripheral motor axons. Its association with either DCC or some UNC5 receptors will lead to axon attraction or repulsion, respectively. Binding to UNC5C might cause dissociation of UNC5C from polymerized TUBB3 in microtubules and thereby lead to increased microtubule dynamics and axon repulsion (PubMed:28483977). Involved in dorsal root ganglion axon projection towards the spinal cord (PubMed:28483977). It also serves as a survival factor via its association with its receptors which prevent the initiation of apoptosis. Involved in tumorigenesis by regulating apoptosis (PubMed:15343335).

Netrin-1 also known as NTN1 or Netrin-1 protein is a guidance cue important for cell migrations. It falls under the netrin proteins and has an approximate mass of 70 kDa. This protein is mainly expressed in the central nervous system especially during development but also appears in certain developing tissues and organs. Netrin-1 interacts with various receptors influencing axon guidance and cell motility.
Netrin proteins play key roles in directing neuronal and vascular development. Netrin-1 influences axon pathfinding by attracting or repelling growth cones and also participates in angiogenesis. It does not usually form a stable complex but functions through interactions with distinct receptor types like DCC Neogenin and UNC5 homologue proteins. These interactions determine the direction of cellular movement and position within tissues.
Netrin-1 integrates within signaling pathways such as the MAPK and PI3K/AKT pathways. It influences neuron migration and blood vessel formation within these pathways. Netrin receptors like DCC and UNC5B are important in transducing netrin-1 signals. These pathways involve intricate signaling cascades modulating cell survival growth and trajectory establishing netrin-1 as significant in neurodevelopmental processes.
Researchers link netrin-1 to neurodegenerative diseases and cancer metastasis. Abnormal levels of netrin-1 can associate with disorders like Alzheimer's disease and various cancers. In cancer netrin-1 interaction with its receptors might influence tumour cell survival and invasion resembling mechanisms found in neuronal cells. Understanding the role of netrin-1 in these conditions could uncover valuable therapeutic targets potentially involving DCC and UNC5 homologues among other associated molecules.

Anti-Netrin 1 antibody [EPR5428] ab126729 was shown to react with NTN1 in wild-type MCF7 cell culture media in Western blot with loss of signal observed in culture media from NTN1 knockout cell line Human NTN1 knockout MCF7 cell line ab323856. Wild-type MCF7 and NTN1 knockout cell culture medium was subjected to SDS-PAGE. Membranes were blocked in 5% milk in TBST for 1 hr before incubation with Anti-Netrin 1 antibody [EPR5428] ab126729 overnight at 4 °C at a 1/2000 dilution. Blots were incubated with secondary antibodies at 0.2 µg/mL before imaging.
