Supplementary information

Netrin-3 also known as NTN3 is a member of the netrin family of proteins and plays a role in guiding axonal growth during neural development. This protein has an approximate molecular mass of 65 kDa. Scientists have observed Netrin-3 expression mainly in the nervous system including the central and peripheral nervous tissues. In addition to nervous tissues it shows presence in various other tissues such as the pancreas and the lungs indicating a wider range of functional implications.
Biological function summary

Netrin-3 helps with cellular movement and positioning in the developing nervous system. It is not part of a larger protein complex but acts through interactions with its receptors like DCC (Deleted in Colorectal Cancer) and neogenin near the cell surface. These interactions facilitate not only guidance of axons but also influence cell adhesion processes impacting how cells navigate during development.

Pathways

Netrin-3 is actively involved in at least two critical pathways: the axon guidance pathway and the cell adhesion pathway. In the axon guidance pathway Netrin-3 interacts with proteins such as DCC to direct the movements of neuronal axons. In the cell adhesion pathway it collaborates with neogenin to support cell attachment and migration essential for structuring tissues during development. These interactions show that Netrin-3 plays important roles in shaping cellular architecture.

Abnormal function or expression of Netrin-3 connects to neurological disorders like schizophrenia and neurodevelopmental disorders. The relationship with proteins like DCC and neogenin can influence the onset of these conditions. Alterations in Netrin-3 levels can disrupt the normal pathways impacting neural development and leading to these complex disorders. Understanding its role opens avenues for potential therapeutic strategies targeting these pathways.
