Supplementary information

Neurobeachin-like 1 (NBEAL1) is a protein involved in several cellular activities. Also known as KIAA1566 it has a molecular mass of approximately 327 kDa. NBEAL1 is expressed in a variety of human tissues including brain and testes. It contains several domains such as the BEACH coiled-coil and WD40 domains which are important for its intracellular functions. Through these domains NBEAL1 mechanistically engages in protein-protein interactions and possibly influences vesicular trafficking and signal transduction within the cell.
Biological function summary

NBEAL1 helps in the regulation of synaptic activity and neuronal growth. It functions within larger protein complexes that are involved in the synaptic vesicle cycle influencing neurotransmitter release and synaptic plasticity. The BEACH domain in NBEAL1 is critical for its involvement in endosomal and lysosomal trafficking. This function can modulate cellular homeostasis impacting various neuronal processes essential for interpreting external and internal signals.

Pathways

Several cellular pathways involve the roles of NBEAL1 notably in neuronal function. It is an essential participant in synaptic vesicle endocytosis and recycling. In this context it interacts with proteins like CALM and AP2 which are integral to the formation and reconnection of synaptic vesicles within nerve terminals. This association places NBEAL1 within the larger framework of neurotransmission and maintenance of synaptic integrity.

Alterations in NBEAL1 function relate to neurological conditions such as autism spectrum disorders and neurodegenerative diseases. Studies suggest that NBEAL1 interaction with proteins like TSC1 and TSC2 may contribute to these conditions possibly through disrupted cellular signaling and synaptic abnormalities. Understanding NBEAL1 interactions opens avenues for exploring therapeutic potential in neurodevelopmental and neurodegenerative disorders.
