Supplementary information

The NEUROD4 gene encodes a neuronal differentiation factor known as Neurogenic Differentiation 4 or bHLH (basic helix-loop-helix family member E33). This protein plays a critical role in neurogenesis and is closely related to the NeuroD family. NeuroD4 weighs approximately 42 kDa and expresses mainly in the developing central nervous system particularly in regions associated with sensory neuron development.
Biological function summary

Neurogenic Differentiation 4 functions as a transcription factor regulating gene expression during neural development. It participates in forming transcriptional complexes with other bHLH proteins facilitating neuronal differentiation and proliferation. By binding to E-box sequences in the DNA NEUROD4 influences the transcription of genes important for neuronal identity and precursory cell maturation.

Pathways

The NeuroD4 protein integrates into neurodevelopmental pathways including the Notch signaling and the Wnt signaling pathways. These pathways are pivotal for the control of cell fate decisions and proliferation in the nervous system. NeuroD4 interacts with other proteins within these pathways such as Notch intracellular domain proteins and TCF/LEF transcription factors further influencing the regulation of specific gene sets important for neural development.

Neurogenic Differentiation 4 associates with neurodevelopmental disorders like autism spectrum disorder and neuronal structure abnormalities such as retinal degenerations. Mutations or dysregulation of NEUROD4 can disrupt normal neural differentiation processes leading to these conditions. Connections to related proteins in these disorders include interactions with Onecut transcription factors relevant in developing retinal and other neural tissues.
