Tubulin is the major constituent of microtubules, a cylinder consisting of laterally associated linear protofilaments composed of alpha- and beta-tubulin heterodimers (PubMed:34996871). Microtubules grow by the addition of GTP-tubulin dimers to the microtubule end, where a stabilizing cap forms (PubMed:34996871). Below the cap, tubulin dimers are in GDP-bound state, owing to GTPase activity of alpha-tubulin (PubMed:34996871). TUBB3 plays a critical role in proper axon guidance and maintenance (PubMed:20074521). Binding of NTN1/Netrin-1 to its receptor UNC5C might cause dissociation of UNC5C from polymerized TUBB3 in microtubules and thereby lead to increased microtubule dynamics and axon repulsion (PubMed:28483977). Plays a role in dorsal root ganglion axon projection towards the spinal cord (PubMed:28483977).

Neuron-specific beta III tubulin also known as TUBB3 is a microtubule-associated protein with a molecular mass of approximately 50 kDa. This protein is expressed mostly in neurons but you may also find it in certain non-neuronal cells under specific conditions. As an important component of the cytoskeleton beta III tubulin collaborates with other tubulins to form microtubules essential for maintaining the structural integrity and transport functions within nerve cells.
Beta III tubulin maintains neuron stability and aids in processes like axonal guidance and dendritic growth. It does not usually form part of a larger complex but interacts dynamically with other microtubule-associated proteins to ensure proper neuron development and function. During neuronal differentiation beta III tubulin is often used as a marker due to its specific expression pattern in neurons compared to other tubulin isoforms.
Beta III tubulin plays an important role in the microtubule-based processes that are critical for the neurodevelopmental pathways. It interacts closely with proteins like Tau and MAP2 regulators of microtubule stability. These interactions fall within the larger scope of the axonal transport pathway a major pathway that involves other proteins such as kinesins and dyneins which drive cellular cargoes along the microtubules.
Beta III tubulin mutations relate to conditions like congenital fibrosis of the extraocular muscles (CFEOM) and neurological diseases. These mutations often disturb microtubule dynamics leading to impaired neuronal signaling and transport. The protein interacts with components like kinesin family motor proteins that contribute to axonal transport highlighting its significance in maintaining neural function and its potential in understanding neural-related diseases.
