Neuropeptide S also called NPS functions as a signaling molecule in the brain influencing arousal alertness and anxiety modulation. This peptide consists of 20 amino acids with an approximate mass of 2 kDa. Expression of NPS occurs mainly in distinct brainstem areas including the locus coeruleus. Neuropeptide S interacts with its specific receptor NPSR1 to exert its effects. Its receptor has widespread distribution not only in the central nervous system but also peripheral tissues.
Biological function summary

Neuropeptide S impacts the brain's response to stress and emotion regulation. It does not form part of a complex rather it binds directly with its receptor to initiate signaling cascades. NPS can enhance arousal and promote wakefulness which affects the sleep-wake cycle and stress response. The activation of NPSR1 by Neuropeptide S also plays a role in reducing anxiety-like behaviors. Through these functions Neuropeptide S influences various behavioral and physiological processes.
